Around her, kids scurry around preparing the ship for flight. Luna’s friend Suzy shouts: Action Stations! We have bacteriophage on the radar. Luna smashes the glass on a red fire-axe style case. Luna and Simon excitedly cut up the swirling strands of DNA that are now flooding the room. Simon is curious: how does the bacteria make sure not to cut up its own DNA? Luna smiles in response. She points out that the scissors are programmed to target a specific sequence found in the phage’s DNA. They can’t cut anything else. Luna hangs the photo and associated scissors on the wall, alongside a huge array of different coloured DNA and scissors. Luna explains that it’s a constant game of attack and counter-attack as bacteria and phage evolve to survive. Luna: Even though we're separated by billions of years of evolution, if you look closely there are a lot of similarities between human and bacterial immune systems. Simon: Maybe I should go into a dormancy period to get better… Luna’s nose tickles and she lets out a huge sneeze. Simon crosses his arms smugly. Tales From the Lab Luna and Simon’s mum gives them a tour of her lab: As researchers around the world discover more and more bacteria, we are finding an astounding variety of different bacterial immune systems. Different bacteria have different combinations of immune systems. Some have a variety of defences, while others are more vulnerable. They stand in front a huge table map showing all the different sites across the UK where research is going on. We see one scientist: I'm crawling through genome data to map patterns of defences. By seeing which ones appear together, we can investigate whether they work together too. We see another, pipetting liquid into test-tube: Here, we're testing thousands of strains for their resistance to different phages. Page 15. Another scientist looks at a screen with a cell lit up with glow in the dark proteins: We want t o understand how cells know when they're infected. And how do defences know when to activate? Here we're using bright fluorescent labels which light up when different defences come online. A scientist build a molecular model: There's still so much we don’t know about the molecular machinery of bacterial immunity. We use x-rays to build an atomic-scale model of the different components to understand how they interact. We see two similar agar plates with bacteria - they put different strains to the test to see which combinations of defences help them survive different conditions. Which strains survive a barrage of phages, and which ones succeed when things are easier? Meanwhile, bacterial immunity can also block the transfer of DNA between bacteria, which can be bad for bacteria that gain useful genes this way. Luna’s mum concludes: It’s a lot of work, but worth it. We’re figuring out how bacterial immunity affects both disease-causing microbes and the beneficial bugs in our microbiome.
